Abstract

The development of gastroretentive drug delivery systems aims to enhance the bioavailability and therapeutic efficacy of medications with narrow absorption windows in the gastrointestinal tract. This study focuses on the formulation and optimization of innovative gastroretentive floating matrix tablets of Alfuzosin Hydrochloride, a drug commonly used in the management of benign prostatic hyperplasia. The floating matrix tablets were prepared using a direct compression method with various polymers to achieve the desired floating properties and controlled drug release. Sodium bicarbonate and citric acid were incorporated as gas-generating agents to ensure buoyancy. A comprehensive evaluation of the prepared formulations was conducted, including in vitro buoyancy studies, drug release profiles, and kinetic modelling. The optimization process utilized a central composite design (CCD) to determine the ideal combination of polymer concentrations and gas-generating agents. Results indicated that the optimized formulation exhibited a floating lag time of less than 2 minutes and sustained drug release over 24 hours, following a zero-order kinetic model. The floating matrix tablets maintained their integrity and buoyancy throughout the release period, demonstrating potential for improved gastric retention and enhanced bioavailability of Alfuzosin Hydrochloride. the optimized gastroretentive floating matrix tablet formulation of Alfuzosin Hydrochloride shows promise as an effective delivery system, offering prolonged gastric residence time and controlled drug release. The formulation 5 can potentially improve patient compliance and therapeutic outcomes in the treatment of benign prostatic hyperplasia. Further in vivo studies are recommended to confirm these findings.
